Description
The CRISPRaTest™ assay measures the transcriptional activator efficacy of any of the several dCas9-activator variants used for the CRISPRa system in any mammalian cell system. Whether you are using dCas9-VP64, dCas-Rta, Cellecta’s dCas9-VPH, or interested in comparing these with a newer Cas9-hybrid activator, this kit will provide you with a quantitative measurement of how well the activator works. Cellecta's Functional Cas9/dCas9 Fluorescent Reporter Assays work differently than Cellecta's Cas9 Essential Gene Assays (i.e., the CRISPRtest kits). While readout for both types of Cas9 assays relies on the the disappearance of a fluorescent marker to assess Cas9 activity, the Essential-Gene Cas9 relies on cell death induced by disruption of a gene required for viability, whereas, with the CRISPR-test Fluorescent Reporter Assays, the sgRNA directly interferes with expression of one of the reporter genes. As a result, the Cellecta Functional Cas9/dCas9 Fluorescent Reporter Assays can be used with any mammalian cell system. Each kit provides enough reagents to measure 5-10 cell lines
